Either its wild-type allele has been entered into the list of genes (mal3) to produce MAL3/mal3 or mal3/MAL3, or "option/space" has been entered into the gene list to produce "MAL3/ " or " /MAL3". (However, "Sort" would move the later to the beginning of the list.)Another option to enter a diploid strain is by specifying its parents; the respective button "Mate Strains" appears on choosing "diploid". The parent strains can be selected by entering their numbers in the strain collection (the parent strains are displayed in the field "Gene Info" after each input), or they can be chosen from lists of search results by clicking them in the list with the "Shmoo" cursor. This option is especially helpful when the mating partners have been chosen by searching them in the collection. Both parent strains must be haploid; strains with identical mating types are accepted after check-back. Plasmids are automatically trans- ferred from the parents; their genes are combined to pairs as good as possible. For that purpose, the genes of one parent are searched for in the set of genes of the other; genes without an obvious pendant are entered as "gene/ " or " /gene". To complement this entry to "GENE/gene, it has to be deleted and the combination of genes entered via selection from the list of genes. The possibility to "mate" still insures that no parent gene is forgotten, and helps complementing the data by presenting all genes, making written notes unnecessary. Genes already present in the strain's list when choosing "Mate strains" are quenched. Alternate names, the origin and other commentary can only be found by “Search commentary”, which can also find strains by their name or a contained plasmid (e.g., if it is known only in part; the search with ”Search for strain name” or “Search for plasmid” is faster and eliminates false positives, e.g., if the commentary mentions the plasmid searched for in a context like "cured of YCp50").Caution: If the name of a strain begins with the name of a gene (e.g., URA3-2C) it will be considered when genes are searched for. If the name of the strain or the commentary contains "/" followed by the name of a gene (e.g., /ura3), this may be considered when genes are searched for in diploid strains.When searching for strains, either haploid strains or diploid strains can be searched for.The search for diploid strains can be restricted to strains with a complete "heterozygous" mating type (a/ª or ª/a), or to diploid strains able to mate (a/a or ª/ª); or any diploid yeast, even with partly or totally unknown mating type (e.g., /a), can be searched. With the last option, the search takes a multiple of the other options.Search for haploid strains: - The desired mating type is chosen or switched off (set to   neutral). Genes are first clicked in the list of genes. <text>This stack was created to allow a clear overview of our yeast strain collec- tion, which is stored in boxes of 10x10 strains at -70°C. The stack has been optimized for the organism and storage form; by using the commentary field for any text, it might be adapted for other kinds of collections. The adapta- tion to other formats than 10x10, however, would be complicated.Clicking the texts below the buttons displays short infos of the functions. More detailed information about the function and usage can be viewed on the respective card.Each box of the collection is represented by a list of strains, which holds the data of each strain in a line. To modify the (write protected) list of strains (add, delete, copy strains, modify or complete strain data), choose the posi- tion of the respective strain in its box. For some tasks, a modification card is opened. On entry of the data, modifications are performed automatically (spaces and returns are changed to option/space, ASCII 202; tabs are inser- ted), that are essential for searching and display. The list of strains must not be deprotected and modified directly.On entering strain characteristics, mutant alleles are selected from a self- produced list of genes by clicking, and the mating type is specified with radio buttons. Plasmids, alternate names of the strain, its origin and any commen- tary can be entered into text fields. Diploid strains can be entered by choosing the parent strains.Yeast strains can be extracted from the collection using various criteria (name, mating type, genes and alleles, haploid/diploid, plasmid contents, commentary). Excerpts can be assembled by searching strains and assembling them in a result list, which can be modified by deleting or pasting strains. The data can be printed or exported as a text only file (e.g., for a dissertation or publication).In it's (normally hidden) graphic, the "Strain Collection" card holds a button, which is intended for illustration of a button in the Home card leading to this stack. Click with the selection tool or lasso, and confirm "Show card picture?" with "OK". The graphic is displayed on the front of the freezer and can be selected, copied and pasted to the Home card.On setup of a new collection, all necessary cards and resources are trans- ferred, the list of genes only if requested, and an empty list for box 1 is produced. Transfer of the two printing templates needs some human help (Who knows how to automate this? The problem is that I know of no scripting command à la "Copy Report", and the script waits after opening the report window, until that is closed again). The to and fro between the new and old stack are run by the script; the user has to copy, then paste the two reports and each time close the report windows. Besides the check-back "Save Report", it is possible that requests like "Cancel Printing?" (with only one choice) or "Altered paper size, save adapted report?" (Yes or no, whatever you prefer) appear.</text>
